Apolipoprotein E (apoE) plays an important role in the metabolism and redistribution of lipoproteins and cholesterol.
ApoE plays a major role in detoxifying HNE and the neuroprotective effects of its isoforms may help explain the relationship between apoE genotype and the susceptibility to neurodegenerative diseases.

Increasing evidence indicates that there are hormonal reductions of estrogen and androgen levels in aged men and women. These hormonal reductions might be risk factors for cognitive impairments and the development of Alzheimer's disease (AD) and Atherosclerosis. Studies have revealed that aged people show improved cognition after treatments with sex steroids. Therefore, ongoing clinical AD trials have been designed to evaluate the potential benefits of estrogen therapy in women and testosterone therapy in men.

Apolipoprotein E (apoE) plays an important role in the metabolism and redistribution of lipoproteins and cholesterol.

Data published in the Journal of Neurochemistry refers to Apolipoprotein E as having a mechanism for neuroprotective effects. This data suggests that apoE plays three important roles in detoxifying a lipid (fat) peroxidation product called HNE or 4-Hydroxynonenal which has been found to covalently bind to proteins on cysteine residues in the spinal cords of people with ALS



The three major human apoE isoforms, apoE2, apoE3, and apoE4, differ in their effects on AD and Atherosclerosis risk and pathology.

E2/E4 - Intermediate protection from Alzheimer's Disease, however elevated risk for Atherosclerosis.
E3/E3 - Normal type (moderate protection from both Alzheimer's Disease and Atherosclerosis).
E3/E4 - Increased risk for Alzheimer's Disease.
E4/E4 - Severe risk for Alzheimer's Disease.
